Ex vivo myelin water and DTI measurements of SKP-SC transplanted cell therapy in contused rat spinal cord: correlation with histology

چکیده
Introduction Schwann cells differentiated from skin-derived precursors (SKP-SCs) have been shown to promote histological and functional recovery in a contusion model of rat spinal cord injury [1]. Cellular bands of SKP-SCs bridge the lesion site which promote axon growth ensheathed by peripheral (Schwann cell) myelin of endogenous as well as transplant origins. In this work, we assess the myelin content and axonal integrity in an ex vivo specimen of contused thoracic cord treated with SKP-SCs by measuring myelin water fraction (MWF) [2] and longitudinal diffusivity (Dlong), and compare them to corresponding histological measures.

Skin-derived precursors generate myelinating Schwann cells that promote remyelination and functional recovery after contusion spinal cord injury.
Transplantation of exogenous cells is one approach to spinal cord repair that could potentially enhance the growth and myelination of endogenous axons. Here, we asked whether skin-derived precursors (SKPs), a neural crest-like precursor that can be isolated and expanded from mammalian skin, could be used to repair the injured rat spinal cord. To ask this question, we isolated and expanded genet...

Kozlowski Prostate Centre, Vancouver General Hospital, Vancouver, BC, Canada, University of British Columbia, Vancouver, BC, Canada Introduction Changes in the amount of myelin water in spinal cord (SC) occur at the time of spinal cord injury and during the subsequent deterioration and recovery phases. Tracking the progression of myelin wa...

Introduction A wide array of processes occurs after spinal cord injury, including white matter demyelination and proximal and distal Wallerian degeneration. The demyelination and interruption of the axonal tract ultimately leads to the functional loss seen in spinal cord injuries. Therefore it is important to have a non-invasive technique that is able to track myelin in spinal cords. There are ...
متن کاملSchwann cell but not olfactory ensheathing glia transplants improve hindlimb locomotor performance in the moderately contused adult rat thoracic spinal cord.
Cultured adult rat Schwann cells (SCs) or olfactory ensheathing glia (OEG), or both, were transplanted in the adult Fischer rat thoracic (T9) spinal cord 1 week after a moderate contusion (10 gm, 12.5 mm, NYU impactor). Rats received either a total of 2 x 10(6) cells suspended in culture medium or culture medium only (controls). At 12 weeks after injury, all grafted animals exhibited diminished...

A major goal of Schwann cell (SC) transplantation for spinal cord injury (SCI) is to fill the injury site to create a bridge for regenerating axons. However, transplantation of peripheral nerve SCs requires an invasive biopsy, which may result in nerve damage and donor site morbidity. SCs derived from multipotent stem cells found in skin dermis (SKP-SCs) are a promising alternative. Regardless ...
